Western Cape Health and Wellness said the roll-out formed part of efforts to reduce new HIV infections in the province.
The initial allocation of 29,000 doses was directed to the City of Cape Town metro.
The drug would be made available at 22 health facilities located in communities with the highest HIV burden, after the province acquired an additional 24,000 HIV infections over the past year.
On the first day of treatment, patients will receive two injections and two oral tablets at the clinic, followed by two more tablets to take at home the next day.
Lenacapavir began working on the third day through a slow-release mechanism that provided protection for up to 28 weeks.
Patients are required to then return to the clinic one month later for a follow-up assessment
